Characterization of an Unusual α‐Oxoamine Synthase Off‐Loading Domain from a Cyanobacterial Type I Fatty Acid Synthase

چکیده
A new fatty acid synthase off-loading domain. Fatty (FAS) usually releases its products by hydrolysis or transesterification. Here, an α-oxoamine domain is identified in a cyanobacterial type I FAS. This the acyl chain through condensation with L-serine, offering novel pathway to α-amino ketones that share similarities sphingolipid precursors. More information can be found Research Article A. E. Characterization of cyclopropane fatty-acid synthase from Sterculia foetida.
Cyclopropane synthase from Sterculia foetida developing seeds catalyzes the addition of a methylene group from S-adenosylmethionine to the cis double bond of oleic acid (Bao, X., Katz, S., Pollard, M., and Ohlrogge, J. (2002) Proc. Natl. Acad. Sci. U. S. A. 99, 7172-7177). To understand this enzyme better, differential expression in leaf and seed tissues, protein properties, and substrate prefe...
متن کاملCharacterization of flavone synthase I from rice.
Flavones are synthesized from flavanones through the action of flavone synthases (FNSs). There are two FNSs, FNS I and II. FNS I is a soluble dioxygenase present in members of the Apiaceae family and FNS II is a membrane bound cytochrome P450 enzyme that has been identified in numerous plant species. In this study, we cloned OsFNS I-1 from rice by RTPCR, expressed it in E. coli, and purified th...

Therapies that target tumor metabolism represent a new horizon in anticancer therapies. In particular, cancer cells are dependent on the generation of lipids, which are essential for cell membrane synthesis, modification of proteins, and localization of many oncogenic signal transduction enzymes. Because fatty acids are the building blocks of these important lipids, fatty acid synthase (FASN) e...

While a deep understanding of the fungal and mammalian multi-enzyme type I fatty-acid synthases (FAS I) has been achieved in recent years, the bacterial FAS I family, which is narrowly distributed within the Actinomycetales genera Mycobacterium, Corynebacterium and Nocardia, is still poorly understood. This is of particular relevance for two reasons: (i) although homologous to fungal FAS I, cry...
متن کاملFIRE (fatty acid synthase insulin-responsive element) and ICE (inverted CCAAT element) regulate fatty acid synthase.
Introduction Fatty acid synthase (FAS) plays a central role in de novo lipogenesis in mammals and birds by catalysing the reaction in the conversion of acetyl-CoA and malonyl-CoA into palmitate. FAS activity is not known to be regulated by allosteric effectors or covalent modifications. However, FAS concentration is exquisitely sensitive to nutritional, hormonal and developmental states [ 1-31....
